I’ve been researching peptides for over ten years.
In that time, I’ve watched a lot of people mess up their PT-141 dosing.
Too much and you’re dealing with nausea and flushing that ruins the whole experience.
Too little and nothing happens.
Getting the dose right matters. And I’m going to walk you through exactly how to do it.

What Is PT-141 and How Does It Work?
PT-141 is also called bremelanotide.
It works differently than anything else people use for sexual function.
It doesn’t act on blood flow. It acts on the brain.
Specifically, it binds to melanocortin receptors in your central nervous system. That triggers a cascade that releases dopamine and oxytocin. Both of those play a direct role in sexual arousal and desire.
So instead of forcing a mechanical response, PT-141 actually increases your desire at the neurological level.
That’s why it works for both men and women. And that’s why dosing it correctly is so important. You’re working with brain chemistry, not just blood flow.
What Is the Right PT-141 Dosage?
There’s no single universal dose. It depends on your body and your goals.
That said, here’s what the research and the real-world data actually show:
For men: Most people start at 1mg and work up to 1.75mg. Some go to 2mg. I wouldn’t go higher than that on the first few runs.
For women: Research typically shows lower doses work well. Starting at 0.75mg to 1mg is common. Women tend to be more sensitive to PT-141 than men.
Timing: Take it 30 to 60 minutes before you want the effect. The peptide needs time to cross into the CNS and do its thing.
Frequency: Don’t use it every day. Most protocols call for use no more than once every 3 to 4 days. Your receptors need time to reset.
How to Use the PT-141 Dosage Calculator
A dosage calculator is a useful starting point if you’re new to PT-141.
The idea is simple. You put in your personal data (body weight, age, gender, health history) and it generates a personalized starting dose.
You can access a PT-141 dosage calculator at peptidecalc.com.
Here’s how to use it correctly:
Step 1: Enter Your Body Weight Accurately
Your body weight is one of the biggest factors in dosing.
Heavier individuals typically need a slightly higher dose to feel the same effect. Don’t round up or estimate here. Use your actual weight.
Step 2: Enter Your Age and Gender
These matter more than people expect.
Hormonal profiles change with age, and men and women respond to PT-141 differently. Getting this right changes the recommendation significantly.
Step 3: Include Any Relevant Health Conditions or Medications
This is where most people skip steps and get into trouble.
If you’re on blood pressure medications, antidepressants, or anything that affects cardiovascular function, that changes how PT-141 interacts in your system.
Be honest here. The calculator can only be as accurate as the information you give it.
Step 4: Review the Output and Start Low
Whatever the calculator suggests, I recommend starting at the lower end of that range.
You can always increase next time. You can’t undo a dose that’s too high.

Why Personalized PT-141 Dosing Matters
I’ve talked to hundreds of people in my Skool community about their PT-141 experiences.
The ones who struggle almost always made the same mistake. They used a flat dose they read somewhere without adjusting for their own situation.
The people who got great results dialed in their dose over a few sessions. They paid attention to how their body responded and adjusted from there.
That’s the whole point of personalized dosing. You’re not just following a formula. You’re learning what your body actually responds to.

PT-141 Side Effects to Know About
I’ll be straight with you. PT-141 does have some side effects. Knowing them ahead of time takes away most of the worry.
Nausea: The most common one. Usually happens in the first 30 to 90 minutes. Tends to go away as you dial in the right dose. Starting low helps a lot here.
Flushing: Some people get a warm, flushed feeling, especially in the face. Same deal. Usually dose-dependent.
Headache: Less common. Staying hydrated before and after helps.
Skin darkening: PT-141 activates melanocortin receptors, which also play a role in pigmentation. Occasional use at normal doses doesn’t usually cause noticeable changes. This is more of a concern with very high or frequent doses.
None of these are reasons to avoid PT-141. They’re reasons to dose it correctly and give your body time to adjust.
Does PT-141 Require a Prescription?
It depends on how you’re using it.
Vyleesi is the FDA-approved pharmaceutical version of bremelanotide. That requires a prescription and is specifically approved for premenopausal women with hypoactive sexual desire disorder (HSDD).
PT-141 sold by research peptide vendors is a different category. It’s sold as a research chemical for laboratory use, not as a prescription drug.
Either way, if you have existing health conditions or are on other medications, talking to a doctor before starting any peptide protocol is the right move.
Does PT-141 Interact with Other Medications?
This is worth paying attention to.
PT-141 can interact with medications that affect blood pressure. When it activates the melanocortin system, it can cause a temporary blood pressure increase in some people.
If you’re on antihypertensives or anything cardiovascular, talk to your doctor first.
It may also interact with drugs that affect the central nervous system given that’s where PT-141 is primarily working. Antidepressants are worth discussing with a healthcare provider before adding PT-141.
Where to Buy PT-141
I source my peptides from BioEdge Research Labs.
They provide third-party lab testing on their peptides so you actually know what you’re getting. That matters more than most people realize. There’s a lot of garbage out there being sold as research-grade.

Frequently Asked Questions About PT-141 Dosage
What is the standard PT-141 starting dose?
Most people start at 1mg for men and 0.75mg for women. Go low on your first run and see how your body responds before increasing.
How long before sex should I take PT-141?
30 to 60 minutes is the standard window. Some people find 45 minutes to be the sweet spot. It takes time to cross the blood-brain barrier and activate the melanocortin receptors.
How often can I use PT-141?
No more than once every 3 to 4 days. Using it more frequently doesn’t increase the effect. It just increases side effects and receptor desensitization.
Can women use PT-141?
Yes. PT-141 is actually one of the few peptides with clinical research specifically in women. The FDA-approved version (Vyleesi) was approved specifically for women. Women typically use slightly lower doses than men and often respond well to 0.75mg to 1mg.
What should I do if I miss a dose?
PT-141 is taken as needed, not on a daily schedule. If you planned to use it and didn’t, just wait until next time. There’s no ongoing protocol to maintain.
Can I stack PT-141 with other peptides?
Some people do. The most common stacks include BPC-157 for general wellbeing or certain hormone optimization peptides. That said, if you’re new to PT-141, run it solo first so you understand how it affects you before adding anything.
Does PT-141 increase testosterone?
Not directly. It works through the melanocortin system, not the HPG axis. It enhances desire and arousal neurologically, not by raising testosterone levels.
